Colm Meaney began working as an actor in Dublin. In the 1970s, he performed with the leftist 7:84 Theater Company, a troupe presenting plays for "working-class audiences", dealing with "political, social and cultural issues of inequality, prejudice and oppression." He has also performed with the Irish National Theatre.

His first film was an action picture about Neo-Nazis, Omega Syndrome with Ken Wahl and Xander Berkeley. He was briefly a regular on the soap One Life to Live, but his recurring role on the Star Trek shows in the 1990s made him a familiar face. He also had major roles in the art films The Snapper, The Commitments, and The Road to Wellville, and played the tart-tongued FBI agent in Con Air with Nicolas Cage.

On Star Trek: The Next Generation, he played Miles O'Brien, the transporter technician, and on Star Trek: Deep Space Nine he was promoted to Chief of Operations.

Since Star Trek, he has appeared in The Boys from County Clare, Bad Apple, Layer Cake. His performance as Harry in How Harry Became A Tree won Meaney a Best Actor award at the 2003 Newport Beach Film Festival.
